Methodology
The LDI method uses as a substrate L-lactate buffered at a pH of 9.4. Lactate dehydrogenase oxidizes the substrate in the presence of NAD+ to yield pyruvate and NADH which absorbs light at 340 nm. Lactate dehydrogenase activity is measured as a rate reaction at 340/700 nm, proportional to the amount of lactate dehydrogenase in the sample. Siemens Dimension Vista.
