Collection Requirements

Specimen must be drawn at POB or any patient service center with centrifuge and refrigeration capabilities.

Keep on ice until processed.

Recommended that patient is fasting and at complete rest.

Primary Collection Container

K Oxlate and Na Fluoride – Grey Top

Transport

Plasma on ice (Inpatient)

Plasma Frozen (Outpatient)

Preferred Transport Temperature

Frozen

Processing

Specimen must be received for centrifugation within 15 minutes of draw time and immediately separated from cells.

Stability

Ambient: 15 minutes

Refrigerated: 15 minutes

Frozen: 7 days

Minimum Testing Volume

0.2 mL

Reference Range

0.5 – 2.2 mmol/L

Methodology

Rabbit muscle lactic dehydrogenase (LDH) catalyzes the oxidation of L-lactate to pyruvate with simultaneous reduction of nicotinamide adenine dinucleotide (NAD). One mole of NAD is converted to one mole of NADH for each mole (equivalent) of lactate present. The absorbance due to NADH is directly proportional to the lactate concentration and is measured using a two-filter (340 – 383 nm) end point technique. Siemens Dimension Vista.

Performed

Sunday – Saturday

Reported

As completed

CPT Codes

83605

Unacceptable Conditions

Failure to deliver to processing center within 15 min.

Refrigerated in excess of 2 hours prior to testing.
